SunScript^® One Step RT-PCR Kit is an easy and reliable system designed for fast RT-PCR reactions using a novel Reverse Transcriptase.

A key advantage of 4BB™ SunScript^® is the elevated temperature for performing the RT step thus allowing higher specificity of the reaction, and a higher likelihood in obtaining reaction products for difficult targets. Moreover, the excellent kinetic performance of the 4BB™ SunScript^® Reverse Transcriptase allows for speed optimization of the protocol. The kit contains all the components needed to perform reverse transcription PCR amplification. The Reverse Transcriptase step can be completed in as little as five minutes.

Supply:
4BB™ SunScript^® One Step RT-PCR Kit is supplied in a single box, with sufficient amount for either 25 or 100 reactions. Contents are provided in clear, screw cap tubes with color coded lids as referenced under ‘Contents’ at the end of this document.

Shipping and Storage:
This product is shipped on dry ice. On receipt, it should be stored immediately at -20ºC in a non frost free (constant temperature) freezer. If stored correctly, the product can be kept for at least six months after shipping without displaying any reduction in performance. For longer periods of time, store the kit at -80ºC. Avoid frequent freeze thaw cycles. Avoid longer exposure of kit contents to light.

Technical Specifications:
The system uses an enzyme master mix containing SunScript^® Reverse Transcriptase RNaseH, a high quality HotStart Taq Polymerase, RNase inhibitor, and a 2X buffer containing dNTPs, salts, stabilizers and the double stranded DNA binding dye.

4BB™ SunScript^® Reverse Transcriptase RNaseH has an increased thermostability, which enables the reverse transcription step to be performed at temperatures up to 75ºC if needed (the recommended temperature for the RT reaction is 60ºC for most samples depending on the melting temperature of the primers). These features make this kit the best choice for amplifying a broad range of mRNAs, including those with complicated secondary structures or high GC content. The reverse transcription reaction can be effectively performed while Taq Polymerase remains inactive during this step due to blocking antibodies. The inactivation / activation step at 95ºC provides an automatic ‘hot start’ for the Taq Polymerase to amplify the reverse transcribed cDNA.

The reaction mixes contain proprietary formulations that have been optimized for both the reverse transcription and the PCR reaction, including dNTPs, MgCl₂ and stabilizers.

Each batch of 4BB™ SunScript^® One Step RT-PCR Kit and individual components are tested against predetermined specifications to ensure consistent product quality.
