Z-Leu-Leu-Glu-AMC is a fluorogenic substrate for the caspase-like activity of the proteasome. The activity may be stimulated in the presence of Mg2+ ions and rapidly inactivated by N-acetylimidazole.

Activity is quantified by release of free fluorescent 7-amino-4-methylcoumarin (AMC) which excites at 360-380 nm and emits at 440-460 nm.
