Alkyne-dATP can be easily incorporated into DNA by PCR using polymerases like Pwo, Deep Vent exo- or KOD XL. The resulting stable and modified (we recommend a modification of 1-10% of the corresponding natural base only) PCR fragment is now ready for further functionalization with a.g. dyes, biomolecules, oligos, etc.
