cPK Assay Kit

Description: Useful for determining the activity of Cytosolic Protamine Kinase (cPK) or related protein serine/threonine kinase. Can be used to evaluate the effect of a test substance on cPK activity or to identify potential new physiological substrates of the enzyme. Kit provides for 50 assays. Assay is based on the reaction of cPK with protamine sulfate and [γ-32P]ATP. Kit includes directional insert, and contains solutions of MgATP, protamine sulfate, assay and dilution buffer, and purified cPK sample. Store solutions at -70° C. Avoid repeated thawing. [γ-32P]ATP and trichloroacetic acid are not provided with kit.

Background: Cytosolic Protamine Kinase (cPK) is a distinct insulin-stimulated protein kinase which phosphorylates eIF4E (GLO118-001, GLO118-005), hnRNP protein A1 and numerous other cellular proteins. cPK is itself regulated by reversible phosphorylation. Phosphorylation by Myelin Basic Protein Kinases MBPK-1 (GLO113-100) and MBPK-2 (GLO114-100) activates cPK. Dephosphorylation by protein phosphatases PP2A1 (GLO130-001, GLO130-005), PP2A2 (GLO131-001, GLO131-005) and PP2AC (GLO132-001) inactivates the kinase.
