PAK Detection Kit

Description: The PAK Detection Kit includes a 200 μg aliquot of affinity-purified Anti-PAK (GLO152-100) and a sample of highly purified PAK1 (GLO110-001). The PAK1 aliquot is provided in SDS sample buffer ready for use in five applications as positive control in Western blots.

The antibody was produced in rabbits to a synthetic peptide corresponding to residues 484-502 of human PAK1. The polyclonal antibody is useful for Western blotting, for example, with 30 μg HEK293 cell extracts at 1:1000 dilution of the antibody. In addition to PAK1, the antibody displays reactivity with PAK2.

Background: The p21-GTPase-activated protein kinases (PAK’s) represent a family of enzymes that are involved in regulating cell morphology and cytoskeleton organization, cell motility, apoptosis, neuronal functions, cell cycle progression, viral replication and numerous other signal transduction processes. PAK’s have been found to interact with protein phosphatase 2A (PP2A, GLO130-GLO132), ribosomal protein S6 kinase, Mixed Lineage Kinase (MLK2), the SH3-SH2 adapter Nck and other regulatory proteins. Physiological targets of PAK include p38 MAP kinase, JNK, Raf1 and Synapsin I. The catalytic domains of PAK family members are produced in apoptotic cells via a caspase-dependent pathway. Unlike the native enzymes, the catalytic domains do not require Rac or Cdc42 (GLO128-005, GLO128-025, GLO128-050) to undergo autophosphorylation and activation.
